Proper support was only added in Linux kernel 3.1 or greater. Use a Linux distribution with at least this kernel for best results.
No issues at all with the Asus K53SJ, works great.
Discussion
Update to ASUS K53SJ and Linux.
Now I'am running OpenSuSE 12.1 on this machine. 3D Support is now available and all the desktop effects are running well. Some other 3D programs are also working. Bumblebee is no longer needed. The card reader still needs the additional driver mentioned above. The touchpad supports the multi finger gestures now. Up to three fingers are now recognized and supported. The webcam works fine with x64 programs. Programs using the i586 libraries are still have the upside down picture problem.
OpenSuSE 12.1 works fine with this laptop.
Skype needs some tweaks but is also working. Suspend to ram (closing the lid) and waking up works. Till now I couldn't find any flaws.
I was looking for a solution on the card reader problem and I found one.
At the following address you can download a driver for the card reader. (Realtek 0bda:0139)

After reboot the card reader is recognized and works fine.
The driver name is rts5139 an will be loaded automatically on system start up. No additional setup is required.

Got a ASUS K53SJ and here some information about running OpenSuSE 11.4 on it.
Graphics : The K53SJ uses Optimus Technology and primary the build in Intel Chipset is used. The NVIDIA Chipset can be used with “bumblebee” and optirun. Limited support for desktop effects.
Sound : Works out of the box
DVD : You can play and burn DVD out of the box
WLAN : works out of the box
LAN : Not tested yet.
Touchpad : Works with some limitations. Tapping works but there is no scroll area or scrolling with two fingers.
Card Reader : not working - but im trying to find a solution
Web-Cam : Working - but the image is upside down.
USB Ports : working out of the box
External VGA : not tested
Headphone : Working
OpenSuSE 11.4 (x64) runs smooth and stable.
